Proteomic Analysis of Drosophila suzukii (Diptera: Drosophilidae) by Fumigant and Low-temperature Combination Treatment

Abstract

The spotted wing Drosophila, Drosophila suzukii is an insect pest that causes significant yield losses to berry crops, cherries, grapes, and other fruits. Combining phosphine (PH3) and ethylformate (EF), which are well-known fumigants, with low temperatures increases the insecticidal effect of D. suzukii. In this study, apoptosis analysis using TUNEL assay kit and protein changes in D. suzukii through two-dimensional electrophoresis (2DE) and MALDI-TOF/TOF were analyzed. As a result, apoptosis pattern tended to be stronger in each fumigant or low-temperature alone treatment group than in the untreated group, and in the combination treatment group of low-temperature and fumigant than alone. In 2DE analysis, about 800 spots were detected and 42 differentially expressed protein spots were observed in D. suzukii. Several proteins, including cathepsin D and heat shock protein 83 were overexpressed in the fumigant + low temperature treatment. These results can be an important indicator for confirming physiological changes in the synergistic effect of insecticidal effects of D. suzukii by fumigant and low-temperature complex treatment.
