Transcriptomic Analysis of Gene Expression Changes in Thrips nigropilosus Uzel under Low-Temperature Exposure

Abstract

Thrips nigropilosus Uzel is a polyphagous thrips species causing economic damage to medicinal and horticultural crops, yet its molecular responses to low temperature remain unclear. Here, we analyzed the transcriptome of adults exposed to 4°C for 24 h (LOW) compared with a control at 25°C (CON). Illumina NovaSeq sequencing generated 7.38 × 108 paired-end reads, and de novo assembly produced 124,562 unigenes (N50 = 948 bp; average length = 611.4 bp). Functional annotation showed that 37.2% of unigenes matched at least one public database, with Gene Ontology terms mainly related to metabolic process and binding. Differential expression analysis identified 1,881 DEGs (860 up- and 1,021 down-regulated in LOW), indicating overall transcriptional suppression under cold stress. Cold-related genes, including acyl-CoA desaturase, cuticle protein, cytochrome P450, glutathione S-transferase, trehalose transporter, juvenile hormone esterase, and lipase, were significantly regulated, whereas vitellogenin was strongly down-regulated, suggesting a shift from reproduction to survival. RT-qPCR confirmed expression patterns consistent with RNA-seq results. This study provides the first transcriptomic resource characterizing the acute cold exposure response in T. nigropilosus and supports the development of improved thrips management strategies.
